英文摘要	Just like in animals, the essence of fertilization process in plants is the fusion of male and female gametes. Although it is already possible, with the aid of cytological manipulation, to achieve in vitro fusion of isolated egg and sperm cell to form zygote([1]), it remains to be solved whether the fusion of male and female gametes in nature is at random or there exists some kind of ''recognition''([2]). Studies on fertilization in animals showed that the fusion of sperm and egg includes a series of complicated processes. The glycoprotein ZP3 present in the mouse egg's zone pellucida serves as a sperm receptor([3]). Subsequently, the acrosome reaction is triggered, enabling passage of sperm through the extracellular coat. Finally, sperm contact and fuse with egg's plasma membrane. In plants, the picture is so far unclear. In order to explore whether there exists special functional protein on the surface of plant gametes and further study its role in double fertilization, it is necessary, first of all, to obtain a large amount of purified viable sperm cells and egg cells. In a previous note([4]), we have reported the successful mass preparation of viable sperm cells from tricellular pollen of Zea mays. The present note reports further isolation of various organelles from the sperm cells, with sucrose gradient ultracentrifugation. The different gradient fractions where different organelles were distributed were characterized by different marker enzymes.,The gradient layer enriched with plasma membranes was collected, extracted with water-satured phenol.
