The role of down-regulation of CD147 in mouse placenta

英文摘要Preeclampsia (PE) is a systemic and complicated pregnancy disease characterized by the syndrome of hypertension, proteinuria and edema after 20 weeks of gestation, affecting the kidney, heart, liver and brain, but the pathogenesis of PE is still unclear. CD147 is a membrane protein that has been implicated in plenty of pathological and physiological conditions. It is detectable in human blastocysts. Reduced placental CD147 expression is associated with PE. Our previous study shown that the invasion ability of trophoblast is reduced when down-regulated the expression of CD147. To address the effects of CD147 on pregnancy outcomes and its relationship with PE, here, a mouse model with the placenta-specific CD147 down-regulation was established through infection of blastocysts with lentivirus of CD147 shRNA followed by blastocyst transfer into pseudopregnant recipients. Blood pressure was measured every 2 days. Samples including blood, urine and kidney were collected on GD10, GD12, GD15 and GD18. Implantation rate, fetal and placental weight were also recorded. Embryo development was monitored using a dedicated small-animal high-resolution ultrasound imaging system. The result showed that resorption rate significantly increased in mice with placenta-specific down-regulation of CD147, and placental and fetal weights significantly decreased on GD15 and GD18. Blood pressure increased beginning at GD12 with higher albumin/creatinine ratio on GD15 and GD18. Embryo development was astricted, displaying fetal growth retardation and placental vascular defects. These results indicate that down-regulation of CD147 resulted in preeclampsia-like symptoms, including hypertension, proteinuria, renal function damage in the animals. In conclusion, a new target for mouse model of PE by down-regulation of CD147 in placenta, which would provide a new target and theory basis for PE diagnosis and treatment.
