Cancer cells fluorescence lifetime imaging based on aptamer functionalized core/shell ZnSe:Mn/ZnS quantum dots

英文摘要Compared with organic dyes, quantum dots (QDs) can be regarded as an excellent candidate for fluorescence lifetime imaging, due to its relatively long fluorescence lifetime, narrow and symmetric emission peak, high photostability, and high photoluminescence quantum yield.It had been well established that the fluorescence of Mn-doped ZnSe nanocrystals at a wavelength around 580 nm was from the transition 4T1to 6A1 of the Mn2+ions in the crystal field of the ZnSe nanocrystals,which always possessed longer lifetime.In this study, we developed a novel strategy to synthesize aptamer (as1411) functionalized water soluble core/shell ZnSe:Mn/ZnS QDs with long fluorescence lifetime(0.2 ms). The Mn-doped ZnSe core was prepared by a nucleation doping approach, and a ZnS shell was grown on ZnSe: Mn2+core throughaddingof Zn2+, 3-mercaptopropionic acid (MPA) and thiourea under appropriate pH conditions. Moreover, as1411 functionalized doped QDs can uniquely identify and track cancer cells in different cells mixture. Furthermore, as-prepared QDs could be used as targeted probes for cancer cells fluorescence lifetime imaging.
