QU FENG ZHI TONG CAPSULE INCREASED MECHANICAL PROPERTIES OF CORTICAL BONE IN OVX-INDUCED RATS

英文摘要Introduction: Qu Feng Zhi Tong capsule (QF) is used for rheumatoid arthritis clinically. It consists of seven herbs, among them Radix dipsaci, Radix angelicaepubescentis and Carthamustinctorius. Ohwi were reported to promote osteogenesis and bone healing traditionaly. Objectives: In this study, we are going to investigate the effect of QF on preventing osteoporosis in vivo. Materials and Methods: Sixty female 6-month-old SD rats were randomly divided into the 6 groups according to body weight. Ten rats were sham operated (Sham) and 50 ones were ovariectomized and then divided into 5 groups, and 10 for each group: three doses of QF (162, 324 and 648 mg/kg/day, i.g. for L-QF, M-QF and H-QF), Estradiol (1 mg/kg/day, i.g.), and OVX (solvent, i.g.). Body weights were recorded every week. All rats were injected subcutaneously with 1% calcein green (10mg/kg) in a time sequence of 13, 12 and 3, 2 days before euthanasia. After treatment for 12 weeks, all of the rats were sacrificed, and the uterus were weighted and embeded, the tibia were collected for high-resolution micro-CT scan and histomorphology analysis, the femur were collected for three-point bending. Results: Body weight in OVX group was significantly higher than that in Sham group at 12 weeks after surgery. All QF groups did not affect the body weights. Compared to the Sham group, the wet uterus weight and epithelial thickness in the OVX group decreased by 80.8%, and estradiol increased 2 folds compared that in the OVX group, while all doses of QF did not affect the wet uterus weight and epithelial thickness. Compared to OVX group, estradiol significantly increased the bone mineral density (BMD), bone volume fraction (BV/TV), trabecular number (Tb.N), trabecular thickness (Tb.th) in the trabecular bone of proximal tibia, while QFs had no effects on trabecular bone. However, H-QF significantly increased the failure load and E-modulus of diaphysis femur. Furthermore, H-QF significantly increased BV/TV of diaphysis tibia, and consistent histomorphometry results showed that H-QF significantly increased the tissue area of cortical bone in tibia. Conclusion: Qu Feng Zhi Tong capsule increased mechanical properties and morphology of cortical bone in OVX-induced rats.
