中文摘要 | sponges, as the best-known source of bioactive marine natural products inmetazoan, play a significant role in marine drug discovery and development. as sessile filter-feeding animals, up to 60% of sponge body could be occupied by associated bacteria. understanding the cellular origin of targeted bioactive compounds from sponges is therefore important for not only revealing the ecological role of these bioactives, but also defining the bioactive production strategy by either sponge cell culture or fermentation of associated bacteria. in our laboratory, two alkaloids hymenialdisine (hd) and debromohymenialdisine (dbh) have been isolated from a sponge axinella sp., which are mapk pathway inhibitors with pharmacological activities of treating osteoarthritis and alzheimer’s disease (zhang et al., 2006). in this study, the cellular localization of these two alkaloids was determined through quantifying these alkaloids in different cell fractions by high performance liquid chromatography (hplc). firstly, using a differential centrifugation method, the dissociated cellswere separated into different fractions according to their sizes. the two bioactive alkaloids were not found in bacteria fraction but in sponge cell fraction obtained from low speed centrifugation. this fraction includes spherulous cells, gray cells and other large mesohyl cells. these cell types were then enriched by a three-step protocol and followed by hplc quantification. compared to the low content in gray cells and large mesohyl cells, dbh and hd content was 10.9% and 6.1% of cell dry weight, respectively in spherulous cells. it suggests that dbh and hd are of sponge origin and located in spherulous cells. in addition, epifluorescence microscopy (salomon et al., 2001) showed that spherulous cells emitted bright red fluorescence under green excitation light, located in tissue paraffin section with a pattern of scattered in mesohyl but abundant in ectosome and excurrent canal surroundings. according to the distribution of spherulous cells, these two alkaloids may act as chemical defense agents against predators. |
