Underivatized amino acids analysis based on ultra performance liquid chromatography coupled to Q-TOF mass spectrometry (UPLC/Q-TOF MS)

中文摘要amino acids play an important role in the metabolism of the human organism. abnormal levels of amino acids suggest that certain metabolism pathway has been disrupted. for instance, phenyl ketonuria will probably appear when phenylalanine can not be transit to tyrosine. amino acids are small molecule with polarity and not or only poorly separated due to weak retention on reversed-phase hplc/uplc column. traditional lc-ms method for amino acids analysis is to enhance retention of amino acids on c18 column by pre-column derivatization or adding ion-paring agent. pre-column derivatization significantly decreases the ionization degree of amino acids which bring on low sensitivity both in positive and negative ion-mode. introducing ion-pairing agents engender ion-suppress which also decrease sensitivity in negative ion-mode. in this study, an acquitytm uplc system (waters, milford, ma, usa) coupled with qtof micro (quadrupole time-of-flight mass spectrometer) was employed. uplc separation was performed on hydrophilic interaction column (1.7μm particle packed). 18 amino acids were chosen as standard samples to study their retention characteristics. through our study, we find 18 amino acids can be detected and better separated on hydrophilic interaction column than on c18 column or other columns. the typical peak widths generated by this method are very satisfied. the reduction in peak width significantly increases analytical sensitivity and concomitant increase in peak capacity, and significantly reduces spectral overlap resulting in superior spectral quality in ms modes. this method for amino acid analysis has great potential in clinic area.
