碳源对巴氏杜氏藻生长及其脂类含量的影响; The Influence of Carbon Resources on the Growth and Lipid Content of Dunaliella bardawil

英文摘要如何提高微藻的生长速度,获取高生物量的产出,同时保持较高的脂类含量是目前微藻生物质能源开发中面临的普遍问题.本实验以巴氏杜氏藻(dunAlIEllA bArdAWIl)突变株Hl-1为对象,研究了不同浓度的碳酸氢钠、葡萄糖、甘油、醋酸钠4种碳源对其生长和脂类含量的影响.结果表明:1)4种碳源对巴氏杜氏藻的生长及生物量的增加均有促进作用,高浓度的碳源对藻生长的促进作用有所减弱,但不抑制生长,其中不同浓度的碳酸氢钠对藻生长的影响差异最明显(最高细胞密度1.18x107 Ml-1,比对照组提高32.8%),而不同浓度的醋酸钠对藻生长的影响最不明显.2)添加低浓度的不同碳源都能相应提高巴氏杜氏藻的生物量,其中生物量最大值为1.24g/l(葡萄糖1MMOl/l),比对照组增加了21.6%,但是随着浓度的升高,生物量有降低趋势.3)不同浓度的甘油、醋酸钠对巴氏杜氏藻细胞的脂类含量差异不显著(P>0.05);一定浓度范围内碳酸氢钠对巴氏杜氏藻细胞的脂类含量差异也不显著(P>0.05),但当浓度升高到12MMOl/l时,与其他浓度组的差异极显著(P<0.01);添加葡萄糖时脂类含量反而更低.但添加碳源能显著促进藻的生长,提高生物量,相应地增加了单位体积内的总脂类含量.4)正交试验中,各试验组的结果差异不大,细胞比生长速率在0.131~0.154d-1间变化,生物量在0.78~1.03g/l间变化,脂类含量在17.5%~21.4%间变化,脂类产率在6.8~11.0Mg/(l·d)间变化.其中细胞比生长速率最高的试验组硝酸钠、尿素、碳酸氢钠、葡萄糖的浓度依次为15,1,6,4MMOl/l.因此很有必要在微藻培养中补充碳源,可提高生长速率,缩短生产周期,又可获得相对较高的脂类含量.; For mass production of bio-fuel,the economic feasibility of microalgal culture greatly depends on the productivity of biomass and lipids.We have studied the effects of sodium bicarbonate,glucose,glycerin and sodium acetate on the growth and lipid content of Dunaliella bardawil HL-1.The results showed as follows:1)Carbon could promote the growth of the D.bardawil HL-1and the promotion effect would diminish in high concentration instead of inhibiting the growth.The effects of concentrations of sodium bicarbonate on growth were significant,with an increase by 32.8%compared to the control.2)The biomass of D.bardawil HL-1could increase with low concentrations of different carbon sources,and when the concentration of glucose was 1mmol/L,the maximum biomass was obtained,which was 21.6% higher than the control.However,with the increase of concentrations,biomass turned to decrease.3)The lipid content of D.bardawil HL-1was slightly effected by glycerol,sodium acetate and sodium bicarbonate at low concentrations,while with concentrations up to 12mmol/L,the difference was extremely significant compared to other cases.4)In the orthogonal test,there was no significant difference between those treatments.The growth rate ranged from 0.131 to 0.154d-1,biomass ranged from 0.78 to 1.03g/L,and lipid content fluctuated between 17.5% and 21.4%.The concentrations of sodium nitrate,urea,sodium bicarbonate,and glucose were 15,1,6,4mmol/L respectively for the highest growth rate.Therefore,it was necessary to add carbon sources to obtain both high growth rate and lipid productivity during Dunaliellaculture.; 福建省科技计划重点项目(2009N0052); 深圳市科技研发资金招研引智项目(ZYD201111080010A); 厦门市海洋经济发展专项(14CZP046HJ20)
