中文摘要	Scygonadin是从锯缘青蟹生殖系统分离获得的一种抗菌肽,我们用Dot-blot检测了该基因在锯缘青蟹幼体(溞状幼体和大眼幼体)和成体锯缘青蟹(雄性和雌性)的鳃、甲壳、眼、血细胞、肝胰脏、心脏、肌肉、甲壳下皮层、胃和生殖系统等10个不同组织器官中的转录表达情况.结果显示,该基因仅在雄性生殖系统中转录表达.进一步分析该基因在性成熟雄性锯缘青蟹和远洋梭子蟹生殖系统中不同部位(精巢、输精管、贮精囊和射精管)的转录表达情况,结果显示,Scygona-din基因主要在性成熟锯缘青蟹的射精管中转录表达.; In this paper,tissue specific transcription of scygonadin gene was further clarified using dot-blot assay.Scygonadin gene is mainly or exclusively expressed in the male reproductive tract of Scylla serrata.The total RNAs were isolated respectively from the different tissues and organs including exoskeleton,subcuticular epithelia,gills,heart,hepatopancreas,stomach,female and male reproductive apparatus,muscle,hemocytes and eyes of the male and female Scylla serrata without artificial challenge.The results showed the scygonadin gene was only expressed in the male reproductive apparatus of Scylla serrata.In order to confirm the tissues-specificity,we dissected the male reproductive apparatus into four parts including testis,seminal vesicle,vas deferens and ejaculatory vesicle.RNAs were prepared from these different tissues and organs respectively,and then analyzed.The results showed that the transcripts were exclusively in the ejaculatory vesicle.The expression of the scygonadin gene in early larval stages of Scylla serrata has also been investigated,the results showed that the gene expression was not detected in those larvae by dot-blot.This observation is consistent with the previous result investigated by RT-PCR assay.; 【作者单位】集美大学水产学院；厦门大学近海海洋环境科学国家重点实验室海洋与环境学院；【作者英文名】HUANG Wen-Shu1,LI Shao-jing2,WANG Ke-jian2(1.Fisheries College,Jimei University,Xiamen 361021,China;2.State Key Laboratory for Marine Environmental Science,College of Oceanography and Environmental Science,Xiamen University,Xiamen 361005,China)
