中文摘要目的:评估热籽感应加温治疗大鼠原位移植性乳腺癌的治疗效果及对免疫功能的影响。方法:89只乳腺皮下接种Walker-256肿瘤细胞的Wistar大鼠随机分为C组(空白对照组,19只),M组(磁场对照组,19只),T组(热籽对照组,19只),H组(加温治疗H_1组,21只;H_2组,21只)。H_1组:45℃常规热疗20分钟;H_2组:50℃~55℃热切除治疗5分钟。加温治疗后即刻,治疗后12 h及24 h各组均处死2只大鼠行常规病理检查及PCNA、HSP70的免疫组化检测。治疗后14天评估大鼠肿瘤体积及观察长期生存。H_1组、H_2组及C组大鼠检测外周血T淋巴细胞亚群CD4~+、CD8~+及IL-2和IFN-γ水平。结果:肿瘤侵入乳腺组织,热疗后肿瘤组织出现坏死及凋亡形态改变。H_1组与H_2组加温治疗后即刻PCNA表达开始减弱,治疗后12h及24h后只有少量残存的肿瘤细胞表达PCNA。H_1组与H_2组加温治疗后即刻HSP70的表达增加。治疗后12h及24h后残存的少许肿瘤细胞表达HSP70。热疗后14天肿瘤体积H_1组、H_2组与3个对照组比较差异有显著性(P<0.05)。H_1组、H_2组与3个对照组比较生存期明显延长(P<0.01),H_1组与H_2组比较无明显差异(P>0.05)。外周血T淋巴细胞亚群CD4~+及IL-2和IFN-γ水平在H_1组、H_2组与C组比较差异有显著性(P<0.05)。结论:两个温度组热籽感应加温治疗对大鼠原位移植性乳腺癌的疗效相当,能提高机体的免疫功能。; Objective:Evaluate the effect of thermoseed inductive heating on mammary orthotopic transplantation tumor in rats and the influence on immunologic function. Methods:Walker-256 tumor cells were inoculated subcutaneously into mammary glands of Wistar rats which were randomly divided into five groups:C Group(control group,19 rats);M Group(magnetic field control group,19 rats);T Group(thermoseed control group,19 rats);H Group(Hyperthermia H_1 groups,21 rats and H2 groups,21 rats).H_1 Group was treatmented conventional hyperthermia,at 45℃for 20 minutes.H_2 Group was treatmented the thermal ablation,at 50℃~55℃for 5 minutes.Immediately,12 h and 24 h after hyperthermia,two rats were sacrificed in each group for routine pathological examination and measuring expression of PCNA and HSP70.Tumor volume was measured 14 days after hyperthermia and long-term survival was observed.T lymphocyte subgroups CD4~+,CD8~+,IL-2 and IFN-γlevels were measured in H_1,H_2 and C Groups. Results:Tumor invadeed mammary tissue in mammary cancer model.After hyperthermia,tumor tissue appeared necrosis and apoptosis.PCNA expression diminished immediately in H_1 group and H_2 group,and 12 h and 24 h later,only small amount of residual tumor cells had PCNA expression.HSP70 expression increased immediately in H_1 group and H_2 group.After 12 h and 24 h small amount of residual tumor cells had HSP70 expression.At the 14 day after hyperthermia, tumor volume of H_1 and H_2 groups was significantly lower than other three groups(P<0.05).Compared with other three groups,the survival period of H_1 and H_2 groups was prolonged significantly(P<0.01),there was no significant difference between H_1 group and H_2 group(p>0.05).CD4+ T lymphocytes as well as levels of IL-2 and IFN-γwere higher in H_1 and H_2 groups than in C group(P<0.05). Conclusions:H_1 Group and H2 Group have equivalent effects in rats mammary tumor orthotopic transplantation and can improve the immunologic function of rats.; 国家自然科学基金10775085; 国家自然科学基金30571799; 清华大学裕元基金~~
