英文摘要Comparative Study of Vaginal Microbiome in Three Closely Related Species of Macaca Ph D. Candidate: Lin Zhu(Cell Biology) Directed by Prof. Yong-Tang Zheng The complex and dynamic host vaginal microbial ecosystem is commonly thought to have been shaped by long-term co-evolutionary processes between the vaginal microbiome and its host and critical to both health and disease of host. Comprehensive understanding the composition and temporal variability of vaginal microbiome remain poorly elucidated due to inherent limitations of culture-dependent methods, low-resolution molecular methods such as clone library, cross-sectional study designs. As high-throughput sequencing-based techniques for microbiome analysis have been developed and used, vaginal microbiome has been revealed as considerably more complex and dynamic. To date, despite the significant advances in understanding of structure and temporal variability of vaginal microbiome in a limited context in terms of host species, resolution of temporal dynamics, and metadata, little is known about two critical questions: whether host genetics contribute to aspects of composition and temporal variability of vaginal microbiome and whether dynamic changes of physiological factors levels contribute to the normal variability in vaginal microbiome through time. Here, we provide a comprehensive catalog of structure and high-resolution dynamics of vaginal microbiome using datasets of 454-pyrosequencing of 16S rRNA genes and metadata including vaginal pH, body temperature, estradiol, and progesterone, covering six cynomolgus, two northern pig-tailed, and six Chinese rhesus macaques over 387 timepoints. Strikingly, we demonstrate host genetics play a predominant role in influencing the structure and temporal dynamics of vaginal microbiome. Next, network analyses reveal the globe and local properties of correlation networks constructed from vaginal microbiome and physiological factors vary widely among three Macaca species, elucidating how the normal variability in vaginal microbiome through time may arise in response to dynamic changes of physiological factors levels. Finally, we uncover phases of the menstrual cycle could influence diversity of vaginal microbiome. Overall, our results highlight the influence of host genetics and physiological factors on complex and dynamic vaginal microbial ecosystem.
